A fresh US drone into Pakistan's northwestern tribal region killed 16 people on Saturday.
The missiles targeted the suspected compound in Dwasarak village, about 40 kilometres (25 miles) west of Wana, the main town in South Waziristan district, a senior Pakistani security official said.
Pakistani media said scores of others were injured in the attack by at least five missile attacks and the death toll was expected to rise.
Two intelligence officials, one in Wana and one based in Peshawar, also confirmed the attack and the death toll.
Officials claimed that suspected fighters belonging to local commander Maulana Halimullah were killed in the attack.
There was no word on the identity of any of the alleged fighters.
Nearly 1,000 people have been killed in more than 100 drone strikes in Pakistan since August 2008, including civilians. The attacks fuel anti-American sentiment in the Muslim country.
